Abstract:
Populus alba var.pyramidalis is one of the important trees in western China.The efficient system of plant regeneration from leaf disc of Populus alba var.pyramidalis was established by the use of selection for basic medium and control of hormone.The effects of auxin,cytokinin,and method of inoculation on the differentiation of adventitious buds from leaves disc were studied.The results showed that auxin was not the important factor and the kinds of cytokinin affected the differentiation rate.The increase of differentiation rate was significant in the medium with relatively low concentration of TDZ.The system of plant regeneration from leaf disc of Populus alba var.pyramidalis consisted of:1/2MS+TDZ 0.005 mg/L+BA 0.25 mg/L+IAA 0.5 mg/L for the differentiation of adventitious buds,MS+BA 0.2 mg/L+IAA 0.2 mg/L for the growth of adventitious buds and 1/2MS for the rooting of adventitious buds.The efficiency of the differentiation of adventitious buds from leaf disc of Populus alba var.pyramidalis reached 100%.
