Abstract:
In order to get Ilex latifolia Thunb.tubeshoots in short time,leaves explant induction callus and tubeshoots production experiments were carried out. Leaves of Ilex latifolia Thunb.were cultured on WPM supplemented with TDZ(0.1~5.0 μmol/L) and IAA(0.1~5.0 μmol/L).Callus production was highest at 5.0 μmol/L IAA,for each TDZ concentration.When these calli were transferred to WPM supplemented with various levels of TDZ(3.0 or 5.0 μmol/L) and IAA(0.0,0.1,1.0,3.0,5.0 μmol/L),the highest number of adventitious shoots(11.2 per callus) formed at 5.0 μmol/L TDZ plus 3.0 μmol/L IAA.A comparison of cytokinin activity showed that the best shoot proliferation was obtained from cultures treated with BA,TDZ and ZT(proliferation times were 3.7,6.0 and 7.4 respectively) at concentrations of 15.0 μmol/L.The longest shoots were produced in cultures grown with BA or 2ip at 3.0 μmol/L(34 mm and 45 mm respectively).
