Abstract:
Pollen viabilities of red alder(Alnus rubra Bong.)were studied in TTC staining and in vitro culture method. The effects of different storage methods on the germination of pollen were also studied. The results showed that TTC method was not suitable for the determination of the pollen viability. The pollen germinating percentage reached the highest after incubated for 8 h on the media supplemented with 10 % sucrose and 0.1 % boric acid. With the extension of storage time, the pollen viability came down. The-20 ℃ environment was the suitable condition for the red alder pollen storage. The pollen thawing effect through tap water was the best in cry opreservation, but pollen storage time was not extended under cry opreservation condition.
