Abstract:
To establish an efficient tissue culture system for rapid propagation of Cyclocarya paliurus,axillary buds from the stem segment were used as explants and cultured on WPM medium. Effects of the different types and concentrations of plant growth regulators on the germination of axillary buds, proliferation of multiple shoots and rooting induction were examined. The results showed that all nine hormone combinations promoted germination of axillary buds to some extent and the most suitable hormone combination was 5 mg/L 6-BA + 1.5 mg/L KT +0.1 mg/L NAA,the germination rate achieved 86.67% at the aforesaid conditions. Putrescine could promote axillary bud germination and shorten the cycle of germination. The combination of 0.3 mg/L TDZ+0.3 mg/L 6-BA was the best treatment for the culture of shoot induction, and the multiplication coefficient could reach 6.78. The treatment of 10mg/L GGR-6+1.0 mg/L IBA had the best effect on rooting induction, and its rooting rate was 25% with superior growth performance.
