摘要:
以火炬松优良家系20692、20615、201036、20639不同球果采收期的含未成熟合子胚的雌配子体为外植体,利用4种针叶树体胚发生专利培养基AL、TX、LP、DS,分别与AL、TX、LP培养基激素组合及无激素组合形成16种诱导培养基,开展不同家系、基本培养基、球果采收期对火炬松体细胞胚胎发生率影响的研究。结果表明:合子胚发育的第2、3、4阶段为最理想外植体,不同家系、基本培养基、激素组合对ESM诱导率及外植体褐化率均存在显著影响,16种组合的培养基中,在LPLP、LPTX、ALTX培养基上可获得较高的体胚诱导率,分别为18%、16%、11%.
