摘要:
<正>采用酶解结合机械震荡法分离出泡桐 (Paulowniafortunei)的生活胚囊。选择新鲜泡桐的胚珠 ,用果胶酶、纤维素酶、离析酶等混和酶液在 2 8~ 30℃下处理 2 .5~ 3h ,并结合机械震荡分离胚囊。经醋酸洋红染色 ,于OlympusAHB 2型显微镜下观察 ,用荧光染料DAPI染色观察生活胚囊内部组成。看到了完整的泡桐生活胚囊的形态。从形态结构上看 ,胚囊内部各成分没有因酶解发生很大的变化
Abstract:
The enzyme shaking technique was used to isolate the viable embryo sac of Paulownia fortunei . Fresh ovules were enzymatically macerated in mixture solution including different content of pictinase(Y 23),cellulase(R 10) and macrozyme(R 10) etc. for 2.5~3?h, at 28~30?℃, under the shaking condition, to isolate the embryo sac. Cytological structure of the whole viable sac was observed under Olympus AHB 2 microscope after aceto carmine staining or DAPI staining.
