<正>以成熟红巴梨果皮的总RNA为模板,根据仁果类果树的f3h基因的保守序列设计引物,利用RTPCR方法从果皮中克隆出花青素生成相关基因f3h基因的全长cDNA,并将该片段连接到pGEM-Teasy载体中。核苷酸序列测定表明,该基因全长1095bp,与苹果的f3h基因同源性高达92%,与矮牵牛相似系数达83%,与拟南芥相似系数达到82%。利用EcoRV和BglIi对重组质粒进行酶切,回收1095bp条带,并将其连接到pBll21载体上。通过抗性筛选和PCR检测,证实了f3h基因已经构建到植物表达载体pBI121上。
Abstract:
Based on climatological standard normals of evaporation and precipitation at an urban area, Nanjing(1960~1990), the variation of two climatic elements in urban forest in Nanjing were analyzed using daily data measured at two meteorological observing stations(an urban site and an open site of the ecological station) from 1988 to 2002. The characteristics of the air moisture were also described by the normals, annual range, month-to-month difference, amplitude of monthly variation and extremum etc. During the past 15 years,the normals of annual evaporation in the urban forest site were 19% less than that in the urban site,mean differences of seasons at two sites were a maximum(25%) in autumn and a minimum(16%) in spring. The normals of annual precipitation in the urban forest site was 4% more than that in the urban site. The mean difference of seasons at two sites was the most(10~) in winter. The result indicated that the urban forest plays an important role in maintaining air moisture near the surface atmospheric layer.
