摘要:
采用抑制性消减杂交(suppression subtractive hybridization,SSH)技术,以杨四瘿螨诱导48、72 h的美洲黑杨无性系为实验方(tester),以未经诱导的美洲黑杨无性系为驱动方(driver)构建杨四瘿螨诱导差异表达的抑制消减cDNA文库,共获得了710个克隆,所建cDNA文库的插入片段主要集中在200~1 000 bp之间。通过序列测定拼接和同源性对比分析,获得了美洲黑杨涉及信号传递、能量代谢、胁迫响应、物质运输等功能的差异基因。
Abstract:
A suppression subtractive hybridization (SSH) cDNA library was constructed with Populus deltoides induced by Tetra lobulifera (Keifer), the leaf was induced for 48 and 72 hours and used as the tester, whereas the induced leaf was employed as driver. Seven hundreds and ten cDNA clones were identified. PCR examination showed that most of the inserted fragments were 200—1 000 bp. The BlastN homology analysis revealed that these genes played varies roles in signal transduction, energy metabolism, response to stress and transportation etc.
