摘要:
【目的】探索蓝莓果实多酚提取物的抗炎作用及其机理。【方法】采用RAW264.7巨噬细胞炎症模型,考察蓝莓果实多酚提取物对一氧化氮合酶(iNOS)基因和环氧合酶-2(COX-2)基因表达以及NO分泌量的影响。【结果】蓝莓果实多酚提取物能抑制脂多糖(LPS)诱导的巨噬细胞中NO的分泌,且随着浓度的增加NO分泌量逐渐降低; 蓝莓果实多酚提取物下调了iNOS和COX-2基因的表达; 参试的5个蓝莓品种果实多酚提取物的抗炎活性有差异。【结论】蓝莓果实多酚提取物能够抑制LPS诱导的RAW264.7巨噬细胞炎症反应,减少NO的分泌,下调被炎症因子激活的iNOS和COX-2基因的表达,从而缓解和抑制炎症的发生和发展。
Abstract:
【Objective】 This study was performed to reveal the anti-inflammatory activity and mechanism of polyphenol extracts from blueberry fruit. 【Method】 The macrophage model was used to investigate the effects of polyphenols extracted from blueberry fruit on the inducible nitric oxide synthase(iNOS)gene and the cyclooxygenase-2(COX-2)gene expression and the NO secretion. 【Result】Blueberry polyphenol extracts significantly decreased NO secretion of lipopolysaccharide(LPS)-induced RAW264.7 macrophages, and higher concentrations of the polyphenol extract resulted in lower NO secretion. Blueberry polyphenol extracts lowered the expression of iNOS and COX-2 genes. The anti-inflammatory activity of polyphenol extracts from five blueberry cultivars was different in this test.【Conclusion】Blueberry polyphenol extract has anti-inflammatory activity by reducing NO secretion and inhibiting the expression of iNOS and COX-2 genes, thus alleviating the occurrence and development of the inflammation of LPS-induced RAW264.7 macrophages.
