摘要: <正>防止黑松外植体及其愈伤组织褐变的研究表明 ,取黑松的成熟胚做外植体诱导愈伤组织的成功率比新梢要高 ;暗培养能显著减轻褐变的发生 ;培养基中附加抗氧化剂Vc和吸附剂活性炭均不能有效抑制继代培养愈伤组织的褐变。选择适宜的培养基类型 ( 1 /2MS)、配合适当的生长调节物质种类及浓度 ,能有效地防止愈伤组织的褐变 ,提高增殖倍数。

Abstract: The methods to preventing the explant and its calli from browning in the processes of vitro propagation of Pinus thunhergii were stu died.The result showed that mature embryo of P.thunhergii is a better explan t than new shoot for its lower browning death rate.Culturing at dark is a necess ary condition for avoiding browning.Adding Vc and active carbon cannot overcome the browning of the calli.Selecting suitable medium (1/2MS) and using proper ho rmones at certain concentrations could prevent browning and increase growth effe ctively.
