野百合与金佛山百合的核型分析及45S rDNA 在其染色体上的物理定位

The karyotype analysis and physical localization of 45S rDNA on the chromosomes of Lilium brownii and L.jinftishanense

摘要: 采用涂片法和45S rDNA荧光原位杂交技术,对野百合(Lilium brownii)和金佛山百合(L.jinftishanense)进行了核型分析和45S rDNA在其染色体上的物理定位。核型分析结果表明:野百合的核型为3B,金佛山百合核型为3A; 野百合的核型公式为2n=2x=24=4m+8st(4SAT)+12t,金佛山百合的核型公式为2n=2x=24=4m+8st+12t; 野百合和金佛山百合的核不对称系数分别为82.12%和82.65%。荧光原位杂交结果表明:野百合有6个45S rDNA位点,它们分别位于3、6、9号染色体上; 金佛山百合有8个45S rDNA位点,它们分别位于1、3、6、8号染色体上。

Abstract: The karyotype analysis and physical localization of 45S rDNA on the chromosomes of Lilium brownii and L.jinftishanense were studied by using the smear methods and the technology of 45S rDNA fluorescence in situ hybridization. The result of karyotype analysis indicated that the karyotype of L.brownii was 3B, the karyotype of L.jinftishanense was 3A; The karyotype formula was 2n=2x=24=4m+8st(4SAT)+12t for L.brownii, 2n=2x=24=4m+8st+12t for L.jinftishanense; As.k was 82.12% and 82.65%respectively. The result of fluorescence in situ hybridization showed that L.brownii had 6 loci of 45S rDNA distributed on chromosome No.3, No.6 and No.9 and L.jinftishanense had 8 on No.1, No.3, No.6 and No.8, respectively.
