桂花及其近缘种倍性和基因组大小分析

Study on ploidy and genome sizes of Osmanthus fragrans and its related species

摘要:

【目的】 解决桂花(Osmanthus fragrans)及其近缘种[柊树(O. heterophyllus)、齿叶木犀(O. × fortunei)和华东木犀(O. cooperi)]在分类处理上存在的较多争议,为桂花及其近缘种的分类研究提供新的依据。【方法】 以水稻品种‘日本晴’(Oryza sativa ‘Nipponbare’)为参照样品,通过流式细胞术对14个桂花代表品种及其3个近缘种的倍性及基因组大小进行分析比对研究。【结果】 四季桂品种群基因组大小为777.24 Mb,金桂品种群为760.85 Mb,银桂品种群为739.06 Mb,丹桂品种群为717.29 Mb。其中‘四季’(O. fragrans ‘Siji’)的基因组最大,为903.14 Mb,‘雄黄’(O. fragrans ‘Xionghuang’)、‘金球’(O. fragrans ‘Jinqiu’)和‘云田彩’(O. fragrans ‘Yuntian Cai’)基因组较大,为801.15~832.79 Mb,超出各品种群的平均值;近缘种华东木犀基因组大小为817.19 Mb,比其他近缘种略高,桂花基因组大小为755.24 Mb,柊树为769.50 Mb,齿叶木犀的基因组最小,为729.11 Mb。【结论】 14个桂花代表品种及其3个近缘种都为二倍体;华东木犀的基因组大小和桂花品种‘雄黄’、‘金球’和‘云田彩’比较接近,都为800 Mb左右,初步表明华东木犀与桂花各品种间具有较为近缘的关系。

Abstract:

【Objective】 This study aimed to provide new cytological evidence for the classification of Osmanthus fragrans and its related species (O. fortunei, O. heterophyllus and O. cooperi).【Method】 In this study, we used representative samples of Oryza sativa ‘Nipponbare’ to estimate the ploidy and genome sizes of 14 O. fragrans cultivars and three related species by using flow cytometry.【Result】 Average values of the O. fragrans Asiaticus Group, Luteus Group, Albus Group and Aurantiacus Group are 777.24, 760.85, 739.06 and 717.29 Mb, respectively. The largest genome size of the cultivar O. fragrans ‘Siji’ was 903.14 Mb. Genome sizes of O. cooperi and O. heterophyllus are 817.19 and 769.50 Mb, respectively; O. fortunei has the smallest genome size (729.11 Mb).【Conclusion】 Fourteen O. fragrans cultivars and three related species were diploid, but their genome sizes differed. The genome size of O. cooperi was close to that of O. fragrans ‘Xionghuang’, O. fragrans ‘Jinqiu’ and O. fragrans ‘Yuntian Cai.’ These results indicated a close relationship between O. cooperi and O. fragrans.
