摘要

【目的】WRI1是调控植物油脂生物合成的核心转录因子。对杜仲EuWRI1进行全基因组鉴定与生物信息学分析可为后续探究EuWRI1基因在杜仲油脂积累中作用机制提供理论参考。【方法】利用生物信息学方法系统分析杜仲EuWRI1基因家族成员,并通过qRT-PCR分析其在不同组织和种仁不同时期的表达模式。【结果】共鉴定到23个含有2个AP2保守域的EuWRI1（即EuWRI1-1~EuWRI1-23）成员,分为4个亚家族,EuWRI1家族的理化性质差异显著,但基因结构和蛋白质序列高度保守,23个成员随机分布在9条染色体上,主要位于细胞核内,包含2个片段重复事件,且与油料科作物的同源性更高。启动含富含MYB、MYC、光响应等相关作用元件,EuWRI1家族基因表达具有组织和时空特异性。【结论】本研究对EuWRI1家族成员进行全基因组鉴定,明确了其基本特征与表达模式,筛选到EuWRI1-9可能作为调控种仁油脂合成的关键转录因子,为进一步解析杜仲油脂积累的分子机制与生物学功能提供了参考依据。

Abstract

【Objective】WRI1 is a key transcription factor regulating plant oil biosynthesis. Conducting a genome-wide identification and bioinformatics analysis of EuWRI1 in Eucommia ulmoides can provide a theoretical reference for subsequent studies on the mechanism by which EuWRI1 genes influence oil accumulation in Eucommia ulmoides. 【Method】Bioinformatics methods were used to systematically analyze the EuWRI1 gene family members in Eucommia ulmoides, and qRT-PCR was conducted to analyze their expression patterns in different tissues and at various stages of seed development. 【Result】A total of 23 EuWRI1 members containing two AP2 conserved domains ( EuWRI1-1 to EuWRI1-23) were identified, divided into four subfamilies. The physicochemical properties of the EuWRI1 family vary significantly, but the gene structures and protein sequences are highly conserved. The 23 members are randomly distributed on nine chromosomes, mainly located in the nucleus, containing two segmental duplication events, and showing higher homology with oilseed crops. The promoters contain elements related to MYB, MYC, and light responses, and the expression of EuWRI1 family genes exhibits tissue- and temporal-specificity. 【Conclusion】This study performed a genome-wide identification of EuWRI1 family members, clarified their basic characteristics and expression patterns, and identified EuWRI1-9 as a potential key transcription factor regulating seed oil synthesis, providing a reference for further elucidating the molecular mechanisms and biological functions of Eucommia oil accumulation.
