柳杉优良无性系离体培养再生体系研究

Study on regeneration system of Cryptomeria clones in vitro culture

摘要: 以柳杉当年生茎段为外植体,采用正交试验设计, 用离体培养的方法,对6个柳杉不同无性系进行芽增殖及3个无性系进行生根诱导,建立了柳杉组培快繁体系。结果表明:①在DCR 添加6-BA、NAA 不同组合的培养基上,6个柳杉无性系均能诱导出不定芽,但不同无性系芽增殖的能力有差异,其中精1#无性系芽增殖系数最高,达到了5.00。②在柳杉无性系离体生根的诱导过程中,1/2DCR培养基是适宜的基本培养基,并且所试验的3个无性系均在添加0.10 mg/L IBA和0.20 mg/L NAA时生根率最高,可达93.3%。

Abstract: The regeneration system in this paper was established by using soft stem of Cryptomeria as explants. Using orthogonal experimental design, among which six Cryptomeria clones were used for the bud proliferation and three clones were used for rooting induction. Main conclusions were shown as follows:① Adventitious bud could be induced by six different clones on DCR basic medium supplement with different combinations of 6-BA and NAA. But there are differences in bud proliferation ability in the different clones. The bud proliferation coefficient was up to 5.00(Jing 1#).② During the process of rooting culture of Cryptomeria clones, 1/2DCR medium was the optimum basic culture medium. And the rooting rate of three clones which had been experimented all reached the highest when added 0.10 mg/L IBA and 0.20 mg/L NAA. Among them, the rooting rate of 011# clone was up to 93.33%. The research will provide technical support for shortening the breeding cycle of the clones of Cryptomeria.
