刺梨黄酮的半仿生法提取工艺及组分研究

摘要: 【目的】探讨半仿生法提取刺梨(Rosa roxburghii Tratt.)黄酮的最佳工艺条件。【方法】结合响应面分析法,考察乙醇浓度、提取温度、料液比和提取时间对刺梨黄酮提取量的影响。【结果】半仿生法提取刺梨黄酮的最佳工艺条件为:乙醇体积分数80%、料液比1:13.2(质量体积比)、提取温度90 ℃、3次提取时间80-40-40 min,此条件下总黄酮提取量为27.70 mg/g,其抗氧化活性显著高于Tween辅助提取和超声波辅助提取的量。经半制备液相色谱分离得到组分Ⅰ和Ⅱ,其出峰时间和槲皮素与山奈酚标准的色谱峰对应。【结论】采用半仿生法提取刺梨黄酮优于传统的乙醇回流提取法,并结合响应面分析法对提取工艺进行了优化,分离纯化后得到槲皮素和山奈酚两种组分。

Abstract: 【Objective】The optimal conditions for the semi-bionic extraction(SBE)of flavonoids from Rosa roxburghii Tratt. were identified. 【Method】Response surface methodology(RSM)was carried out to investigate the effects of ethanol concentration, extraction temperature, solid-liquid ratio, and extraction time on the yield of flavonoids from Rosa roxburghii Tratt. 【Result】The results showed that the optimal conditions for SBE were 80% ethanol, solid-to-liquid ratio of 1:13.2(mass volume ratio), and extraction at 90 ℃ for 80-40-40 min; the extraction yield of flavonoids under these conditions was 27.70 mg/g. The antioxidant activity was significantly higher than that in the case of Tween auxiliary extraction and ultrasonic-assisted extraction. Component I and Component II were separated by semi-preparing liquid chromatography, and their retention times agreed with those of quercetin and kaempferol. 【Conclusion】SBE is superior to the traditional ethanol extraction, and in combination with RSM, offers better yields of purified quercetin and kaempferol.
