杉木萌芽中内源激素的研究

摘要: <正>本文报道在杉木萌芽中存在的几种天然激素。杉芽的醇溶性部分通过阳离子交换树脂、DEAE纤维素柱与纸层析和薄层层析分离,并用小麦芽鞘法、水稻幼苗法、萝卜子叶法及苋红合成法分别对提取分离的各部分进行了生理效应测定。此外还进行‘厂化学鉴定和光谱测定。结果表明,杉芽中含有三大类生长促进物质——生长素,赤霉素、细胞分裂素。除了生长素的含量较低外,赤霉素和细胞分裂素的含量均较高,尤以细胞分裂素的效应最为突出。 经过多次试验,从硅胶GF_(254)薄层层析分离得到六个明显的荧光暗斑,其洗脱液经紫外光谱测定,观察到其中四个部分的吸收峰为270毫微米,与玉米素的吸收峰相一致。另两个部分的吸收峰为260毫微米,与腺嘌呤的吸收光谱相似。经生物鉴定,它们都有细胞分裂素活性。实验表明,杉芽中含有5—6种细胞分裂素类物质。这类激素与杉木很强的萌蘖性之间的关系,值得进一步研究。 从抑制物质含量来看,纸层析和生物鉴定的结果表明,只有Rt0.8—0.9区段出现较为明显的抑制效应。其纸谱浸提液经紫外光谱测定,在波长280毫微米处表现低弱的吸收峰,是一种不同于ABA的物质。这种物质在杉芽中的含量虽较低, 但具有抑制效应。

Abstract: The purpose of this research is to study the natural hormones in the sprouts of Cunninghamia lanceolata (Lamb.) Hook. Samples were partitioned in solvents and isolated by cation exchange resins and DEAE-cellulose columns. The extracts were purified by paper and thin-layer chromatography and the fractions were verified by means of bioassay, with wheat coleoptile section being adopted for IAA and ABA, rice seedling for GAs, and radish cotyledon for cytokinins respectively. The results showed that there existed in the extracts only a little auxin and growth inhibitors but quite some GAs and cytokinins, with the activities of cytokinins being most prominent.By TLC on silica gel GF254 and using the solvent butanol-isopropanol-ammonia-water (2:8:1:1) for the separation of cytokinins, six dark spots appeared in the UV light. The spots were eluted with 90% ethanol separately. The eluates were examined with a spectrophotometer. The ultraviolet absorptive spectra showed that four eluates had weak absorptive peaks at 270 nm, and the other two eluates showed high peaks at 260 nm, all of them having biological activities of cytokinins. From this experiment we may conclude that the sprouts are rich in cytokinins, which might be an important factor causing the buds to have high vigor for sprouting.
