新疆杨植株再生体系的建立

Establishment of Plant Regeneration System of Populus alba var.pyramidalis

摘要: <正>从培养基筛选、激素调控等方面探索建立新疆杨高效植株再生体系。结果表明,不同种类的细胞分裂素影响叶盘不定芽分化频率,其中较低浓度的TDZ(Thidiazuron)可显著提高叶盘不定芽分化频率,生长素的种类与浓度对叶盘不定芽分化影响不显著。较适合新疆杨不定芽分化的培养基为:1/2MS+TDZ0.005mg/L+BA0.25mg/L+IAA0.5mg/L;壮苗培养基为:MS+BA0.2mg/L+IAA0.2mg/L;生根培养基为:1/2MS。在此条件下,不定芽分化频率可达100%。

Abstract: Populus alba var.pyramidalis is one of the important trees in western China.The efficient system of plant regeneration from leaf disc of Populus alba var.pyramidalis was established by the use of selection for basic medium and control of hormone.The effects of auxin,cytokinin,and method of inoculation on the differentiation of adventitious buds from leaves disc were studied.The results showed that auxin was not the important factor and the kinds of cytokinin affected the differentiation rate.The increase of differentiation rate was significant in the medium with relatively low concentration of TDZ.The system of plant regeneration from leaf disc of Populus alba var.pyramidalis consisted of:1/2MS+TDZ 0.005 mg/L+BA 0.25 mg/L+IAA 0.5 mg/L for the differentiation of adventitious buds,MS+BA 0.2 mg/L+IAA 0.2 mg/L for the growth of adventitious buds and 1/2MS for the rooting of adventitious buds.The efficiency of the differentiation of adventitious buds from leaf disc of Populus alba var.pyramidalis reached 100%.
