The Application of RAPD Markers in Diversity Detection and Cultivars Identification of Osmanthus fragrans

摘要: <正>从100个随机引物中筛选出28个多态性稳定的引物,对34个桂花品种、6个桂花野生居群个体、5个木犀属其他种进行了遗传多样性和亲缘关系的RAPD分析。共检测了231个位点,其中209个位点为多态位点(90.4%)。不同品种群的多态性位点频率差异明显,按大小依次排序为:银桂品种群>四季桂品种群、金桂品种群>丹桂品种群。聚类结果与形态分类结果不完全一致。利用来自引物A11和M18扩增的21条多态性条带构建了桂花品种的DNA指纹图谱,该图谱可以方便地应用于桂花品种的鉴定。不同桂花品种之间存在明显差异。研究结果表明,RAPD技术可用于桂花品种的鉴定和亲缘关系的探讨。

Abstract: Based on 28 Primers which are of stable polymorphism selected from 100 random primers, RAPD polymorphism of 34 sweet osmanthus(Osmanthus fragrans)cultivars, 6 individuals from wild population and 5 other species of Osmanthus, and their relationships were detected. Among a total of 231 fragments observed, 209 fragments were polymorphic (90.4%). There were obvious differentiation among different goup in polymorphism, and the order was as follows: Latifolius Group>Fragrans Group and Thunbergii Group>Auran-tiacus Group. The results of cluster analysis were consistent with morphological classification by and large. A DNA fingerprinting based on 21 bands amplified with All and M18 was constructed and might be used in sweet osmanthus cultivars identification. There were obvious differentiaion among different cultivars . In addition, it is feasible to identify the sweet osmanthus cultivars and to study their relationships of this cultivars by use of the RAPD technique.
