Effects of priming on cell cycle and germination of Pinus massoniana seeds

摘要: 采用流式细胞术研究不同引发处理下马尾松种子胚细胞DNA含量的差异,测定不同引发处理下马尾松种子的发芽率。结果表明:(1)PEG 8000溶液引发处理能促进胚细胞DNA的复制,4倍体细胞的比例增加,其中以20 %PEG 8000溶液引发3~4 d的处理对种子胚细胞DNA合成的促进作用最明显。(2)胚细胞增殖指数最高的引发处理是20 %PEG 8000溶液在15 ℃时引发3~4 d。(3)较高浓度的PEG 8000溶液和较长的引发时间有利于提高马尾松种子发芽率。(4)在一定范围内,胚细胞增殖指数与种子发芽率呈显著正相关。

Abstract: In order to reveal the cytological mechanism of promoting seed germination through seed priming, the differences of DNA content in embryonic cell of Pinus massoniana seeds after different priming treatments were studied by using flow cytometry, and seed germination percentage was also determined. The results showed that: (1) DNA replication in embryonic cell could be enhanced by PEG 8000 solution pretreatment, the ratio of tetraploid cells increased, the best treatment was a regime treated for 3-4 days using 20 % PEG 8000 solution. (2) The best priming treatment for increasing cell proliferation index was a regime treated in 20 % PEG 8000 solution for 3-4 days at 15 ℃. (3) Higher PEG 8000 concentration and longer pretreatment time were benefit for increasing germination percentage. (4) In a certain extent, there was a significant positive correlation between the cell proliferation index and seed germination percentage.
