为提高云芝产胞外多糖的产量,在单因素试验的基础上,应用响应面分析法对云芝菌丝体产胞外多糖的培养条件进行了优化。根据回归分析,确定的较佳工艺条件为:培养时间24 d,装液量676 mL,初始pH=51。利用醇沉淀法和DEAE Sepharose CL-6B凝胶柱层析对云芝胞外多糖进行纯化,产品达到电泳纯。紫外扫描和HPLC的分析结果表明,云芝菌株胞外多糖不含多肽,单糖组成至少含有葡萄糖和木糖。
Abstract
To study the optimization of culture conditions on the extracellular polysaccharides Coriolus versicolor, the response surface methodology (RSM) based on the single factor tests were applied. According to regression analysis, the optimal culture conditions were culture time 24 days, broth content 676 mL, and initial pH 51. In order to get the purified polysaccharide, alcohol precipitation and DEAE Sepharose CL6B purification were used. The polysaccharide was identified the uniform component by PAGE. The HPLC results showed that the components of monosaccharide contain at least glucose and xylose, without peptides.
