摘要

<正>以里氏木霉 (TrichodermareeseiRutC30 )为菌种 ,采用改进的Mandels配方能合成酶活力高达 2 5.5IU/mL的木聚糖酶 ,产酶周期 3d。在酶用量 1 % ,温度 50℃的条件下 ,酶解35g/L的纯木聚糖 4~ 1 0h ,可获得 74.0 8%~ 87.89%的低聚糖 ,此时低聚糖与木糖之比为1 6.76~ 1 2 .30。当用 55cm× 7.5cm的凝胶柱在进样量 60mL ,洗脱速度 80 0mL/h ,柱温为50℃的分离条件下 ,能较好地将聚糖与木低聚糖分开 ,总分离时间仅需 3h。

Abstract

The xylanase production, selective enzymatic hydrolysis xylan and isolation xylo oligosaccharides by polyacrylamide gel column were studied.The higher activity of xylanase,up to 25.5?IU/mL, was synthesized by T.reesei Rut C30 with improved mandels cultivation broth, and the time of xylanase production was 3 days.When enzymatic hydrolysis lasted for 4~10 hours at 35?g/L substrate concentration, 50?℃ ,pH?4.8, and 1% enzyme dosage,the yield of oligosaccharides and the ratio of oligosaccharides to xylose were 74.08%~87.89% and 16.76~12.30. Xylo oligosaccharides was well isolated by 55?cm × 7.5?cm polyacrylamide gel column.The separating conditions were:injection volume,60?mL,column temperature,50?℃,elution flow rate, 800?mL/h. In this conditions, the whole separating time was only 3 hours.
