摘要

对3种消毒剂(3 % H2O2、4 % NaClO、0.1 % HgCl2)在22~25 ℃(室温)和4~8 ℃(低温)下对松材线虫的培养效果进行研究,线虫经消毒剂处理后用无菌水清洗,然后在肉汤培养基中培养。结果表明:(1)该3种消毒剂能在室温和低温条件下5 min之内杀死松材线虫虫体上分离的所有细菌菌株;同时也能在室温和低温条件下30 min杀死松材线虫匀浆液中的全部细菌。(2)3种消毒剂在低温4.5 h或室温25 min的条件下,不能完全灭菌。(3)3 % H2O2杀菌效果较好,其次是4 % NaClO,0.1 % HgCl2效果较差。(4)3种消毒剂处理后松材线虫的存活时间(表示为消毒剂,在低温条件下存活时间(在室温条件下存活时间))分别为3 % H2O2, 23 h(>3.5 h);4 % NaClO, 8.5 h(3 h);0.1 % HgCl2, 3.5 h(1 h)。结果显示松材线虫携带的细菌可能隐藏在消毒剂不易到达的线虫体表,或线虫的表被层具有对抗这些消毒剂保护细菌的作用。
