山樱花基因组大小的测定

The genome size determination of Cerasus serrulata

摘要: 为了测定山樱花基因组的大小,获得适合山樱花裂解的流式细胞术的最适裂解液,优化了樱属植物流式细胞术流程。结果显示,在所对比的6种裂解液中,LB01最适合樱属植物细胞裂解。以大豆(Glycine max (L.)Merrill)为内标,比较待测样品和内参峰值,计算得出山樱花基因组大小为366.67 Mb,C值为0.375 pg。可用于樱属植物的倍性鉴定。

Abstract: The present study optimized the process of flow cytometry(FCM)for Cerasus with the purpose of determining the genome size of Cerasus serrulata and obtaining optimal buffer for its FCM process. The results showed that the LB01 was the best buffer for C. serrulata among the six analyzed buffers. This paper used Clycine max(L.)Merrill was the internal standard to test the genome size of C. serrulata, which was 366.67 Mb with C value 0.375 pg. That present results would lay foundations for ploidy identification of Cerasus serrulata.
