摘要: <正>介绍一种显示刺槐根瘤中侵入线超微结构的电镜制样方法。以透射电镜制样方法为基础,将制好的刺槐根瘤树脂块,在同一部位用超薄切片机制成厚切片和超薄切片。厚切片经脱树脂后,扫描电镜观察;超薄切片经染色后,透射电镜镜检。实验结果表明:用该方法处理的样品,可以获得清晰、完整的刺槐根瘤菌侵入线的超微结构及三维结构图像。

Abstract: This paper introduced a method of specimen preparation for electron microscopy which showed the ultrastructure of the infection threads in the host cell of Robinia pseudoacacia. Based on the specimen preparation of TEM, thick sections and ultrathin sections were made at the same location on nodule embeded by Epon_812. Thick sections were de_resined and coated with gold_palladium for SEM; ultrathin sections were stained for TEM. The result showed specimens prepared by this method could obtain ultrastructrue image and three_dimensional image at the same location while infection threads in the host cell of Robinia pseudoacacia.
