摘要: <正>核型分析是现代细胞学和基因组学的重要基础。运用两种预处理方法对杉木核型进行研究 ,即离体根尖在 0~ 4℃的冰水中处理 60~ 72h和离体根尖先用 0 .0 5%的秋水仙素在 0~ 4℃的低温下处理 1 8~ 2 4h后 ,再用 0~ 4℃的冰水处理 1 8~ 2 4h。结果表明 ,用第一种处理方法所获得的中期染色体较多 ,其分散度较好 ,但染色体有多个“缢痕”或出现断裂现象 ,给着丝点和次缢痕的判断带来了困难 ;两种方法所获得的核型模式图差异不明显 ,但第二种方法基本上克服了第一种方法的不足 ,使得染色体更易观察

Abstract: The karyotype of Chinese fir with two improved methods of preparing metaphase chromosomes spread was studied:(1)root tips were treated with 0~4 ℃ iced water, 60~72 hours;(2)before with 0~ 4 ℃ iced water 18~24 hours, root tips were treated with 0.05% colchicine 18~24 hours. The result indicated that: the number of metaphase chromosomes appeared more and chromosome spreaded well with the first method. However, there existed the phenomenon of breakpoint or several “centromeres” in one chromosome. So it was difficult to judge real centromere. The situation can be improved by the second method. The difference between karyotypes treated with two methods was not significant.
