何首乌野生种质资源的RAPD指纹图谱构建

摘要: <正>以甜樱桃矮化砧木Gisela系品种为试材,对影响试管苗生根的因素进行了研究。结果表明,培养基中无机盐浓度、外源激素水平、种类以及外植体的发育状况等对试管嫩茎生根具有明显影响。取继代次数超过30次、生长旺盛、叶小呈嫩黄绿色的嫩茎,接种在1/4MS添加0.5mg/LIBA的诱导生根培养基上,黑暗处理15d,然后移至光下诱导生根,结果发现生根率高,且根生长快、根量多、移栽易于成活,矮化砧木Gisela5,Gisela6离体生根率可达83.3%和86.9%。

Abstract: The ten wild germplasm resources of Guangxi were collected and their inheritable variety were analyzed by the RAPD. The result showed that the PCR amplification with 40 screened primers were conducted to access and 295 genes were obtained from the PCR Amplification, 175(59.3%of total genes) of which were polymorphic. The genetic variation is lowest between the germplasm resource of Tianlin and the germplasm resource of Xilin,and the genetic variation is biggest between the the germplasm resource of Xilin and the germplasm resource of Guanyang. Five tapes of ten Polygonurn multiloru.mclassifiedbylRAPDwere related with their distribution,and some of them from near distribution can get together. The result by the RAPD would offer to scientific theories for the classification,authenticity and the good breeds selective of the germplasm resources of the P. multilorurn.
