Cloning and Sequencing of a Molluscan Glycosyl Hydrolase Core Gene from the Snail, Ampullaria crossean

摘要: <正>以10mmoI/LCaCl2溶液处理花生幼苗叶片后,将花生幼苗置于培养箱在42℃下培养,定时测定幼叶相关生理指标,并观察幼叶超微结构。结果表明:Ca2+处理能有效抑制高温胁迫对花生幼苗叶绿素的破坏和光合速率的下降,提高和保护了Ca2+-ATPase的活性;Ca2+处理能有效减轻高温胁迫对叶绿体、细胞核等细胞器膜结构的损伤。可以认为,Ca2+处理能提高花生幼苗的耐热性。

Abstract: As a mollusk degrading the plant biomass rapidly, Ampullaria crossean can produce cellulose-hydrolytic enzymes. This brings us a new sight into the biological degradation of the plant biomass. Total RNA was isolated from the stomach of the mollusk Arnpullaria crossean by using polymerase chain reaction, cloning and sequencing techniques, a complementary DNA DY1300 encoding a low molecular mass glycosyl hydrolase has been identified. It contains a 882-nucleotide ORF encoding a 294 amino-acid protein. The percentage of similarity with the cellulase EGX gene sequence from Ampullaria crossean is 89.8%. Amino acid sequence-based classification has revealed that the putative protein belongs to the glycosyl hydrolase family 10(GHF10).
