光质和基因型对青钱柳叶黄酮类化合物积累的影响

Influence of light quality and genetype on flavonoid accumulation in the leaves of Cyclocarya paliurus

摘要: 【目的】通过研究光质对不同基因型青钱柳叶黄酮类化合物积累的影响,为青钱柳叶用人工林定向培育提供理论依据。【方法】以4个家系(四川沐川31号、浙江安吉1号、广西金钟山6号、广西金钟山7号)的青钱柳当年生实生苗为试验材料,在人工气候室控制温度和湿度的条件下,设置LED白光、蓝光、红光和绿光4种光质处理,测定其叶片黄酮类化合物含量(总黄酮、异槲皮苷、山奈酚和槲皮素)和合成途径关键酶(苯丙氨酸解氨酶(PAL)、肉桂酸4-羟化酶(C4H)、4-香豆酰-辅酶A连接酶(4CL)和查尔酮合成酶(CHS))活性。【结果】不同光质和基因型对青钱柳叶黄酮类化合物含量和合成途径酶活性均有显著影响,且两者存在显著的交互作用(P<0.05)。总体来讲,与白光和红光相比,短波段的蓝绿光处理下的青钱柳叶黄酮类化合物含量更高。青钱柳叶总黄酮含量和异槲皮苷含量最高的为蓝光处理下的安吉1号家系,而山奈酚含量和槲皮素含量最高的为蓝光处理下的金钟山家系6号和7号。相关性分析表明,总黄酮含量与黄酮类化合物合成途径酶活性均呈现显著相关。【结论】 光质和基因型显著影响青钱柳叶黄酮类化合物含量和合成酶活性,蓝光更有助于黄酮类化合物的积累,可为青钱柳定向培育林分的光照管理提供参考。

Abstract: 【Objective】 The objective of our study was to determine the effects of light quality and genotype on flavonoid accumulation of Cyclocarya paliurus leaves, and thereby provide some basis for its directional cultivation on plantations. 【Methods】 Using current-year seedlings of four C. paliurus families(Muchuan 31#, Anji 1#, Jinzhongshan 6# and 7#)as materials, four LED light-quality groups(white, blue, red and green light)were set under controlled conditions of temperature and humidity in an artificial climate chamber to determine the effects of light on their leaf flavonoid content(total flavonoids, isoquercitrin, kaempferol and quercetin)and enzymes activities(PAL, C4H, 4CL and CHS)for biosynthesis. 【Results】 Both light quality and genotype significantly affected the leaf flavonoid content and enzymes activities for biosynthesis, and a significant interaction of light quality and genotype was observed(P < 0.05). Overall, compared with white and red light treatments, short-wave blue and green light treatments resulted in much higher leaf flavonoid contents. The highest contents of total flavonoids and isoquercitrin were observed under the blue light treatment for the C. paliurus family of Anji 1#, whereas the highest contents of kaempferol and quercetin were observed under blue light treatment for the C. paliurus families of Jinzhongshan 6# and 7#. Pearson's correlation analysis showed that leaf total flavonoid content was significantly correlated with the enzyme activities for biosynthesis that were studied. 【Conclusion】 The results indicated that leaf flavonoid content and enzyme activities for biosynthesis in C. paliurus were significantly modified by light quality and genotype. Blue light had a better positive effect on flavonoid accumulation in C. paliurus leaves, which would provide a reference for the light management in the directional cultivation on plantations.
