Histopathological changes in the resistant pine trees inoculated by pine wood nematode

摘要: <正>采用超临界CO2技术提取香榧外种皮并分析其化学组分,研究了萃取压力、萃取温度、萃取时问和分离温度等单因素对提取物得率的影响,并通过正交试验确定了最佳的提取工艺条件为:萃取压力15MPa,萃取温度45℃,萃取时问120min,分离温度35℃。在上述条件下,提取物得率17.5%。通过GC-MS分析含量较多的40个化学组分,共鉴定出38个化学成分,发现香榧外种皮超临界c0,提取物中大多为萜类化合物。

Abstract: Bursaphelenchus xylophilus isolate was inoculated with one-year-old resistant and susceptible seedlings of Pinus thunbergii respectively. The pathological variation in cells and tissues of host were studied. The results showed that pathological symptoms and damages of cells in resistant seedlings were less than those in susceptible ones "for a given inoculation site. Considerable pathological symptoms were firstly seen to a susceptible. And they were mainly distributed on resin ducts in cortex and parenchyma around them. Outermost xylem also appeared the symptoms. However, those symptoms just confined to parenchyma in cortex, resin ducts and parcnchyma around them for a resistant. It was suggested that varied developments of histopathology in pine infected by pine wood nematode were closely related to the resistance of the seedlings.
