枸杞中黄酮类化合物纯化工艺的研究

Purification process of total flavonoids in Lycium barbarum L.

摘要: 采用静态吸附法筛选适合纯化枸杞黄酮类化合物的大孔树脂,并优化其动态吸附纯化工艺。静态吸附纯化试验表明:供试4种大孔树脂中,HPD-100对枸杞中黄酮类化合物吸附量最高,解吸率达到82.15 %。动态吸附纯化试验表明:上样浓度为5.0 mg/mL、上样流速为2.0 mL/min、洗脱液乙醇质量分数为50 %、洗脱速度为1.5 mL/min时,枸杞总黄酮的纯度为78.53 %,回收率达71.35 %。

Abstract: The optimum macroporous resins for purification of flavonoids in Lycium barbarum L. were screened by static adsorption method. The purification process of flavonoids in L.barbarum L.was optimized through dynamic adsorption experiment. The static adsorption results showed that macroporous resin HPD100 had a highest adsorption in the four types of macroporous resins, and its adsorption rate reached 82.15 %. The dynamic adsorption results showed that when the injecting velocity, injecting concentration, elution solution concentration and eluting velocity were 2.0 mL/min, 5.0 mg/mL, 50 % ethanol and 1.5 mL/min, respectively. The purity of the total flavonoids achieved 78.53 % and the recovery reached 71.35 %.
