Lycoris aurea is a perennial,bulbiferous plant.Its vegetative growth and reproduction are discrete.To study the leaf development of Lycoris aurea,A cDNA library was constructed from Lycoris aurea leaves.The titer of primary library is 5.6×10~5 pfu/mL,and the titer of amplified library is 6.9×10~9 pfu/mL,in which 96% of the phages were recombinant.Insert sizes ranging from 500 bp to 2 500 bp were obtained.All of the above mentioned accorded with the general requirements of cDNA library construction,which established a good foundation for investigating the leaf development and cloning full-length cDNA of genes related to leaf development of Lycoris aurea with the methodology of both genomics and molecular biology.
