Study on methods for rapid determination of seed viability of Santalum album

Abstract: In order to find the most suitable method for rapid determination of seed viability of sandal (Santalum album), TTC method, indigo dying method as well as I-KI reaction method were studied by comparing with the germination test. The results showed that the best testing condition for sandal seed viability determination was staining 3 h with 0.1% TTC at 40 ℃ under darkness. Significant differences were found among TTC concentration(p<0.001), staining temperature(p=0.006)and the interaction between TTC concentration and staining temperature(p=0.001), great differences also existed among different testing methods(p<0.001). TTC method got the most similar results with the germination rate, and would be the most suitable method for rapid determination of seed viability of sandal, while indigo dying method and I-KI reaction method were not suitable.
