球根花卉花粉萌发过程中糖类物质及相关酶活性的变化

Changes of carbohydrate content and related enzyme activity during pollen germination of two bulb flowers

摘要: 为明确兰州百合和唐菖蒲花粉在离体培养中对外源糖的选择存在显著差异的原因,在定性分析这两种球根花卉成熟花粉糖类物质的基础上,对花粉萌发过程中糖类物质含量和相关酶活性进行了测定。结果发现:两种球根花卉花粉中都含有蔗糖、葡萄糖、果糖等可溶性糖,但仅兰州百合花粉中含有淀粉,且可溶性糖含量较高。以PEG4000+硼酸为培养基进行花粉培养,两种花粉均有较高的萌发率; 唐菖蒲花粉在仅添加PEG的培养基中不能萌发,兰州百合花粉则可以萌发。在花粉萌发过程中淀粉酶活性无显著变化,但兰州百合花粉淀粉酶活性显著高于唐菖蒲花粉,蔗糖酶及乳糖酶变化显著,在花粉培养初期迅速下降。淀粉(兰州百合)、蔗糖及葡萄糖含量的变化也很显著,尤其是葡萄糖。因此,两种花粉在PEG培养基中萌发力的差异,以及对外源糖的选择性都或许与花粉中糖类物质的种类、含量及酶的活性不同有关。

Abstract: Lilium pollen differed with Gladiolus pollens on the dependence of pollen germination and pollen tube growth on sugar sources. To clearly know the mechanism of pollen germination of Lilium and Gladiolus, the content of carbohydrates and related enzyme activity of germinating pollen were determined based on qualitative analysis of pollen carbohydrates. Starch was detected in Lilium pollen, but not in Gladiolus pollen. Contents of sucrose, glucose and fructose in Lilium pollen were significantly higher than that in Gladiolus pollen. In addition, germination rate of both kinds of pollen was higher in PEG4000+H3BO3, and Lilium pollen could germinate in culture medium only containing PEG, but Gladiolus pollen could not. When the pollens were cultured in PEG4000+H3BO3, the changes of amylase activity in germinating pollen were not significant, but the amylase activity of Lilium pollen was higher than that of Gladiolus pollen. The changes of invertase and lactase activity in germinating pollen were more significant, and it decreased rapidly at the beginning of pollen culture. The changes of carbohydrate content were very significant, especially glucose. With the rapid growth of pollen tube, glucose content decreased. The difference of pollen germination rate in PEG culture medium and the dependence of pollen germination on sugar sources between Lilium and Gladiolus pollen may be related to the differences of carbohydrate type, content and enzyme activity in nature and germinating pollen.
