Identification of the true hybrids for Populus deltoides by using SSR markers

Abstract: Since the purity of hybrids affects the quality of the genetic map, identification and selection of hybrids should be done before the genetic map construction. In this study, we screened 11 SSR pairs primer that could be used to track the kinship of the hybrids from a total 300 pairs primer. Subsequently, these primers were used to test 380 offsprings from a hybridization of Populus deltoides. Among which, 372 individuals were revealed to own complementary alleles for each parent, and they were identified as true hybrids for genetic map construction. Whereas the remaining eight individuals, they were observed to own non-parental alleles or extra parental alleles, suggesting these progeny were either resulted from pollen contamination or chromosomal abnormalities. Thus they were excluded from map construction.
