Effects of methyl jasmonic acid on somatic embryogenesis of Liriodendron hybrid

Abstract: 【Object】 In order to further increase the frequency of somatic embryogenesis, the experiment was conducted to investigate the effect of methyl jasmonate(MeJA)on somatic embryogenesis development of Liriodendron hybrid. 【Methods】Based on an established system of somatic embryogenesis of Liriodendron hybrid, embryogenic cells of three genotypes included C138, 233012 and 253010 from Liriodendron chinense×L.tulipifera were cultured in the embryo induction medium with different concentrations of MeJA, and the rates of somatic embryogenesis, somatic embryo maturation, abnormal somatic embryos, and number of regeneration plants were investigated. 【Result】 The different concentrations of MeJA can significantly affect the efficiency of Liriodendron hybrid somatic embryogenesis, and with the increase of MeJA concentration, the rates of somatic embryogenesis and maturation gradually increased, the rate of abnormal embryos decreased gradually. However, when the MeJA concentration exceeds a certain value, the rates of somatic embryogenesis and maturation showed a decreased trend, and the number of abnormal embryos also increased. Of which genotype 233012 was in the first place as well as genotype C138 was higher than 253010. Abscisic acid(ABA)could enhance the above-mentioned effects of MeJA. But the increasing concentration of MeJA restricted the root growth, and ABA could not alter this effect. 【Conclusion】The induction effect of Liriodendron hybrid somatic embryos could be optimal when adding 1 mol/L MeJA and 2 mg/L ABA, the rates of somatic embryogenesis and somatic embryo maturation were relatively high, with the lowest rate of abnormal embryos.
