Callus induction from young embryos of Cyclocarya paliurus

Abstract: Using young embryos of Cyclocarya paliurus as explants, we investigated the effects of embryo development period, basic media, types and the concentrations of plant growth regulators on callus induction of C. paliurus. The results showed that there were obvious differences in the shape of callus among different embryo development periods, and the middle cotyledon embryos were most suitable for callus induction. As a basic media, improved DKW medium was the best selection for young embryo callus induction, of which induction rate could hit to 88.89 % with no browning phenomenon. The medium containing 2,4-D and KT was more appropriate for callus induction. The 2,4-D (0.5 mg/L)+KT (2.0 mg/L) as well as 2,4-D (1.0 mg/L)+KT (1.0 mg/L) were optimal for callus induction of C. paliurus young embryo, and the induction rates were 80.00 % and 78.33 % respectively.
