Effects of several factors on rooting of cutting propagation of ancient Platycladus orientalias and the changes of nutritive material

Abstract:

【Objective】 Improve the rooting rate and establish the wig cutting propagation of ancient Platycladus orientalis. 【Method】 In this study, a total of 3 treatment groups using three different concentrations (200-600 mg/L, 1 000-3 000 mg/L and 4 000-10 000 mg/L) of ABT1, NAA or IBA+NAA (mass ratio 1:1) were used to treat 5-, 100-, 300-, 700-year-oldPlatycladus orientalis cuttings. The rooting process, rooting (rooting rate, rooting number, the longest root length) and the dynamic changes in nutrient content (soluble sugar, soluble starch) were observed during the rooting process of Platycladus orientalis cuttings. 【Result】 The observations revealed that 0-35 days was the formation stage, the 35-65 days was adventitious roots formation stage, and 65-95 days was adventitious roots elongation stage. The southern upper cuttings from the 50-year-old P. orientalis exhibited the best rooting percentage of 40.8%. The rooting rate of the 100-year-old P. orientalis was the highest in June with 20.1%. Low concentration growth regulator treatment of ABT1 (600 mg/L) for 4 h was beneficial to root formation of cuttings from 5-year-old P. orientalis, and a mixture solution of medium concentration IBA+NAA(moss ratio 1:1) treatment is good for rooting of cuttings from 100-, 300- and 700-year-old P. orientalis. Further studies revealed that the optimal mixture solution of IBA+NAA (moss ratio 1:1) of 1 000 mg/L for 1 min could improve the rooting rate of cuttings from the ancient P. orientalis. For all age groups, the content of soluble sugar and starch increased during callus formation stage; the 5-year-old P. orientalis showed the maximum increase. 【Conclusion】 Nutrient content of the branches are relatively higher in summer, which is beneficial for increasing the rooting rate of P. orientalis cuttings.
