Identification of Hybrids of Kosteletzkya virginica Using AFLP Markers

Abstract: Kosteletzkya virginica is a potential halophytic grain crop for consumption and industrial utilization. AFLP markers were used to identify 36 F1 hybrids of two parents. Eighteen primer combinations amplified 1 100 AFLP bands between parent Hyl03 and Hh182,108 (9. 82%) of which were polymorphic. Total 1 182 AFLP bands were amplified between parent Slhy353 and Blho301,148(12. 51%) of which were polymorphic. Primer combination E+AAC/M4-CTC amplified five paternal characteristic bands between Hyl03 and Hh182; 401 bp, 390 bp, 156 bp, 137 bp and 108 bp,whereas Eq-AAG/M+CTG generated seven paternal characteristic bands:457 bp,356 bp,312 bp,251 bp,240 bp,190 bp and 76 bp. All these paternal characteristic bands were found in eight F1 hybrids of Hy103 × Hh182,which were real hybrid. Primer combination E+AAC/M+CTT amplified five paternal characteristic bands between Slhy353 and Blho301:440 bp,393 bp,289 bp,162 bp and 67 bp,whereas Eq-ACA/M+CAT generated four characteristic bands.. 448 bp,360 bp,217 bp and 145 bp. All these paternal characteristic bands were found in 26 of 28 F1 hybrids of S1hy353 × Blho301,and other two F1 hybrids were false hybrids.
