Callus Induction and Plantlets Regeneration from Zygotic Embryos of Cunninghamia lanceolata Hook.

Abstract: Using mature zygotic embryos and immature zygotic embryos as initial explants, the effect of the following factors on induction callus and differentiation adventitious buds.. basal medium, plant hormone and development stages of zygotic embryo were studied. Results indicated that the optimal medium for mature zygotic embryos induction callus was 1/2MS+2mg/L 2,4-D+1.0mg/L 6-BA+1. 0mg/L KT+30 g/L sucrose. This medium can induce soft, transparent callus from immature zygotic embryos of embryo selection or tissue and organ differentiation stages. The induction frequency was 2%~6%. Immature zygotic embryos of tissue and organ differentiation or embryo mature stages induction callus were similar in state with those of mature zygotic embryo. The callus induction frequency was significantly improved with the maturity of immature zygotic embryos. The optimal medium for differentiation adventitious buds was DCR+1.5mg/L 6-BA+1. 5mg/L KT+20 g/L sucrose. Shoots with 1. 5 cm high were transfered to rooting medium and rooting frequency reached 100%.
