Studies on Changes of Proteins in the Leaves of Ginkgo biloba by SDS-PAGE and IFE-SDS-PAGE

Abstract: The techniques of SDS-PAGE, IFE-SDS-PAGE, and Brilliant Blue and CommassieDye were used to study the fractions of Vegetative Storage Proteins (VSPs) and the concentrationof the soluble protein in the leaves of Ginkgo biloba. The results showed that the concentrationof the soluble protein in the leaves of Ginkgo biloba (both from a male tree and afemale tree) was higher during the growing period in one year. But it began to decrease at thelast ten-day of October and decreased to the lowest level at the middle ten-day of November, reflecting nitrogen nutrient transfer from the leaves to the other tissues of a tree when theleaves began to fall down. Based on the SDS-PAGE bands, it was found that VSP 55 kD bandstill could be clearly observed in the lane of the middle of October. While this VSP band wasundetectable in the lane of the beginning of November for a female tree, and in the middle ofNovember for a male tree. The ease of VSP 32kD was very similar to VSP 55kD. Only differencewas that VSP 32kD could not be detected until the beginning of November for a femaletree or for a male tree. With the technique of IFE-SDS-PAGE, the seasonal variations ofVSPS 55kD and 32kD were confirmed, and also it was found that another VSP, 18kD, sharplydecreased in the middle of November for a female tree and decreased in the beginning ofNovember for a male tree.
