Enzymology Characteristics of Immobilized β-glucosidase

Abstract

The enzymology characteristics of immoobilized and free 13-glucosidase were investigated. The optimal pH of immobilized and free 13-glucosidase was 4.5. The optimal temperature of immobihzed β-glucosidase was 60 C, which was 5 C lower than that of free 13 glucosi dase. hs stabilities of pH and storing were obviously higher than that of free β-glucosidasc, but its thermal stability was a little lower than that of free 13-glucosidase. The apparent Km and Km of immobilized and free 13-glucosidase were 0.52 mmol/L and 2.4 mmol/L, respec tively. The immobilized 13 glucosidase was used to hydrolyze 10g/L cellobiose solmion re peatedly, its half life time was about 31days.
