Abstract

Adsorption of xylanases from Trichoderma virides to the insoluble oat- spelt xylan and the microerystalline cellulose (MCC) was investigated. It was found that the Langmuir’ s isotherms were suitable for the adsorption of xylanases to the oat-spelt xylan and the MCC, respectively. The adsorption capacities of the oat-spelt xylan and the MCC for the xylanses were 88.9 and 32.4 U/g. The adsorption affinity of xylanases to the oat-spelt xylan was higher than that to MCC. The adsorption of xylanases to the polysaccharides used could be described as the first order kinetic processes. The rate constant of adsorption to oat-spelt xylan was 35% higher than that to MCC. The electrophoresis pattern and Michaelis’ constant showed that the components bounded by the polysaccharides tested were identical. The results suggested that the oat-spelt xylan was a promising candidate adsorbent in the rapid recovery and purification of xylanases.
