Cryopreservation of excised embryos of Aesculus chinensis seeds

Abstract: Through analyzing the seed viability, relative electrical conductivity and dehydrogenase activity of excised embryos after being cryopreserved (-196 ℃), the feasibility of long-term storage of Aesculus chinensis seeds was studied. The results showed that moisture content (MC) was the main factor affecting the cryopreservation of A.chinensis excised embryos and proper dehydration should be conducted in cryopreservation. The optimal regime of treatments was:57.3 % MC+slow freezing(QF)+quick thawing(QT)+10 % DMSO+10 % sucrose+20 % polyethylene glycol. Through this treatment the viability of cryopreserved excised embryos of A.chinensis was 93 %.
