The chromosome karyotype and selection of preparation materials in Apriona germari (Hope)

Abstract: Apriona germari (Hope) was chosen as a model to investigate karyotype in this paper. Ganglions of larvae, ganglions, salivary glands, testis and ovaries of adults, tissues of embryo were used as experimental materials. The results showed that the optimal materials were 2, 3 days old embryos. The nucleic acid content of embryo was determined. The content was increasing with day age, and peak value appeared at 3, 6, 9 days old embryo. The ratio of RNA/DNA in 3 days old embryo was higher than that in 6 or 9days old embryo. When the nucleic acid peak value appeared firstly, the embryo was the most suitable material for the chromosome research. So the ratio of RNA/DNA in embryo could be used as an index of material choice for making chromosome specimen.
