Effects of hormone and light on in vitro preservation of axillary buds of Pinus densiflora resistant to pine wilt disease

Abstract: In order to preserve axillary buds of the Pinus densiflora with excellent resistance against Bursaphelenchus xylophilus, the effects of different treatments of phytohormones and light on bud preservation were studied. It showed that dark condition was not favorable to in vitro bud preservation at 17 ℃. On the contrary, addition of 1.0 mg/L ABA or 02 mg/L 6BA plus 0.1-0.2 mg/L NAA under lightdark alteration had a positive effect on the bud preservation. In this condition, survival rate of the axillary buds was 94 % or higher and all survived buds proliferated well.
