The karyotype analysis of longiflorum lily and oriental lily

Abstract: Plantlet root tips of longiflorum hyhrids ‘White Fox’ and ‘Lei Shan’, oriental hybrids ‘Crystal Blanca’ and ‘Constansa’ were used as materials to analyze karyotype. The root tips were pretreated with cyclohexamide then fixed in Carnoy’s solution and cell wall was digested with cellulase and pectinase. Finally, spreading method was used for preparing slides. The karyotype analysis result indicated that the karyotype of ‘White Fox’ was 3A, that of others were 3B. The karyotype formula was 2n=2x=24=4m+4st(2SAT)+16t for ‘White Fox’, 2n=2x=24=2m+2sm+10st(2SAT)+10t for ‘Lei Shan’, 2n=2x=24=4m+10st+10t for ‘Crystal Blanca’, 2n=2x=24=4m(1SAT)+10st+10t for ‘Constansa’. Karyotype asymmetry coefficient were 83.11%, 82.78%, 79.75%, 81.22%, respectively.
