Induction techniques of adventitious shoot from callus of Cyclocarya paliurus in vitro

Abstract: Based on the seedlings cultured with matured seeds of Cyclocarya paliurus in vitro, the induction of adventitious shoot from callus of C. paliurus and the effects of basic media and different growth regulators on induction of callus and adventitious shoots were studied with explants of leaves and stem segments of the young seedlings. The results showed that callus of leaves and stem segments of the seedlings could be induced on each basic medium applied in this paper and the highest induction frequency of callus reached 90.00% for stem segment and 86.70% for leaf, respectively. The types and concentrations of hormones had significant effects on callus induction. When 0.5 mg/L TDZ was added, the induction frequency of leaf callus reached 95.00%, while 92.85% induction frequency was achieved for stem segment callus at 0.5 mg/L TDZ added. The greatest induction frequency of adventitious shoots from leaf callus was observed in the medium of WPM+1.0 mg/L 6-BA+0.1 mg/L IBA, which reached to 53.85%. Our results successfully achieved the induction and germination of adventitious shoots from the leaf callus, but the germination rate was still low and more studies should be done in the future.
