Ultrastructure of Pagiophloeus tsushimanus Morimoto antennal sensilla with scanning electron microscope

Abstract: 【Objective】Pagiophloeus tsushimanus Morimoto is a wood-boring beetle that has recently been introduced to China causing serious damage to Cinnamomum camphora in Shanghai City and Fujian Province. Investigating the ultrastructure of the pests' antennal sensilla could elucidate its olfactory behaviors and interactions with its host.【Method】The antennae were submerged in an ultrasonic bath, freeze-dried and sputter-coated with gold. Then, the antennae morphology, the type, number and distribution of antennal sensilla were visualized by using scanning electron microscopy.【Result】The adult antennae were geniculate, seven types including 11 subtypes of sensilla were recorded: sensilla chaetica(Sch Ⅰ-Ⅲ), trichoid sensilla(St Ⅰ-Ⅱ), sensilla basiconica(Sb Ⅰ-Ⅱ), cuticular pore, cylindric sensilla, styloconica sensilla and sensilla furcatea. Sensilla chaetica represent the most abundant type of sensillum, and there were no significant sex-specific differences in length, number, or distribution of the antennal sensilla, except for the lengths of Sch Ⅰ and Sch Ⅱ. However, sensilla basiconica and sensilla furcatea were only found in females.【Conclusion】Studying on the ultrastructure of antennal sensilla of female and male P. tsushimanus provides a theoretical foundation for the development of pheromone-based P. tsushimanus control.
