Study on effects of different storage methods on pollen vitality of Osmanthus fragrans

Abstract: The aim was to research the effects of different storage methods on pollen vitality of Osmanthus fragrans(Thunb.)Lour., and explore the suitable temperature condition for pollen storage of sweet Osmanthus. Taking the pollens of nine sweet osmanthus cultivars of four groups as test material, the pollens were preserved in room temperature, low temperature, freezing and supper-low temperature. The pollen viabilities during different storage time were determined. The results indicated that the pollen germination was well developed in the solid medium composed by sucrose 10%, H3BO3 0.01% and agarose 1%. The germination rate could be calculated after 24 hours cultivating under a constant temperature of 25 ℃. The light and dark showed no significant effect on pollen germination. The ability of pollen germination demonstrateed a significant difference in different storage conditions. Under room temperature(15-20 ℃), O. fragrans pollen could survive after 15-20 days. In low temperature, pollen life can be extended to 50 days. In freezing temperature and supper-low temperature, pollen life can be extended longer, and the most(78%)pollen germination of cultivated O. fragrans showed no significant difference after 100 days. Pollen vitality of O. fragrans decline slower under the condition of freezing temperature and supper-low temperature, it can be preserved for longer.
