DNA EXTRACTING METHODS FROM VEGETATIVE TISSUE OF CONIFER

Abstract: The quality of the isolated DNA is one of the most important facts that influence the result of DNA analysis. Some experiments, such as genomic mapping and population genetics require preparation of large numbers of DNA samples that are correctly identified with respect to the individuals that were the source of the DNA samples. The identity of samples must be maintained, but the samples must be processed quickly and efficiently. Here we describe a simple method to isolate high-quality and identified DNA from vegetative tissue of conifer. The crucial step is to select the suitable material. Different from the common used tissues, whose yielded DNAs are often degraded due to its complicated secondary substance and additive, the tender unwoody terminals are adopted for DNA isoaltion. Here also describes two DNA isolation protocols. According to these protocols, DNA yields and purities were similar for all samples. Highly purified DNA from different conifer species can be obtained following these procedures.
